Wales are expected to play against the Eastern Province Kings ahead of their two Tests against the Springboks this June.
Talks between Saru and the Wales Rugby Union about this clash are already at an advanced stage. All that remains is for the decision to be ratified by Saru's executive council.
The tour match could take place on 10 June at the Nelson Mandela Bay Stadium. Wales will then tackle the Boks in Durban on 14 June, and again in Nelspruit a week later.
Wales are bringing a large squad to South Africa and hope to use the match against the Kings to experiment with some new combinations. The Kings will use this opportunity to prepare for their entry into the Currie Cup Premier Division.
Nelson Mandela Bay Stadium will play host to another big clash on 28 June when the Boks play Scotland.
