The Chiefs scored six tries to beat Wales' second-stringers 40-7 in Hamilton on Tuesday.
It is the first time a Super Rugby franchise has beaten a touring international team. The Chiefs were missing eight All Blacks and another 12 players.
The match was all over by half-time as three tries from scrumhalf Brad Weber, No 8 Tom Sanders and lock Dominic Bird gave the hosts a 21-0 lead. A break from former All Blacks flyhalf Stephen Donald led to the first try and he continued to free up the Chiefs' dangerous backs.
Wales, who had been forced to start Test captain Sam Warburton after Ellis Jenkins pulled out because of injury, finally got on the board early in the second half through replacement hooker Kristian Dacey, but that was followed by the Chiefs' fourth try, from fullback James Lowe.
Chiefs winger Toni Pulu and replacement back Sam McNicol then crossed the line to complete the rout.
Wales will now face the All Blacks in Wellington on Saturday having lost the first Test 39-21 in Auckland.
